  癌症是威脅人類生命的主要疾病之一。對癌細胞發生的成因已有諸多的報導,但對癌症的治療仍在尋求最有效的方法。臨床上放射治療仍是常用的方法之一,但對已經有轉移現象 (Metastasis)的腫瘤則缺乏顯著的效果。因此,若能尋求適當的因子其可增加放射治療的效果 (如增加輻射敏感度) 並同時抑制腫瘤的轉移,相信可對癌症治療提供新的治療途徑。
  肌動蛋白骨架 (Actin Cytoskeleton) 是癌細胞轉移重要的生理組成物之一。小 GTPase Rho 家族蛋白質 (small GTPase Rho protein family),包括Rho,Rac 及 Cdc42,可接受胞外訊息的刺激促進細胞生長,同時形成肌動蛋白細胞骨架供細胞附著,伸展,運動,甚至生長。目前已知有30%的癌細胞與此類蛋白的異變有關。Rho 等蛋白可激活磷酸酶 ROCK 或是 PAK等,使其活化 LIM kinase (1)。LIM kinase 在細胞內唯一已知的基質為Cofilin, 是肌動蛋白去聚合因子家族蛋白的一員 (Actin Depolymerization Factor ),約有21kD的大小,主要功能是加速actin dynamics,幫助細胞的移動及化驅性,而LIM kinase 的功能即在於磷酸化並調節 cofilin 活性 (2)。到目前為止,cofilin 是否直接影響癌細胞生長及入侵轉移仍不清楚。而 Rho-LIM kinase的訊息傳導是否僅透過 cofilin 的活性調節達到細胞變異的目的也需進一步探討。
  本實驗室主要研究興趣於探討cofilin 分子做為基因放射治療的可能性,透過分子影像技術於動物體內追蹤以cofilin 為主的基因轉殖細胞所形成的腫瘤,其輻射敏感度是否如體外試驗一般,且轉移入侵能力也受到抑制。另外,cofilin 與其他分子表現調節的關係也將在此計畫中探討之,包括利用microarray檢測在大量表現cofilin的細胞中其基因表現圖譜的變化。此研究結果將有助於癌症治療及新藥開發的可能性。
